The boys Season 5 will be full Supernatural Meeting after Jared Padalecki and Misha Collins confirmed that they joined the cast of the first.

No one knows. The Hollywood Reporter (THR) has affirmed that they will appear in the invited roles, but there are no indications of how large or small will those roles will be.
However, one thing is safe: the trio will share scenes together. After arriving at Amazon MGM Studios, Thr reporter Rick Porter said Ackles, Padalecki and Collins will appear together on the screen at some point in season 5.
That does not mean that Padalecki and Collins join Ackles to adapt as SUPS, although it is certainly a possibility.
